What is the KAM541 crown wheel and pinion set for?

It's a 4.1:1 ratio crown wheel and pinion for the front differential, long nose diff configuration. The pinion, driven by the propshaft, turns the crown wheel and transmits drive through 90 degrees to the differential carrier and halfshafts.

What's the difference between the long nose and short nose diff versions of this ratio?

The long nose diff (this KAM541) and the short nose Wolf diff use different pinion designs and aren't interchangeable. KAM541 is for the long nose diff fitted to Defenders, Discovery 1 and 2, and Range Rover Classic. If your vehicle uses a Wolf-type short nose diff, you need KAM549 instead at this ratio.

Do front and rear differentials need to match this ratio?

Yes, front and rear must run the same ratio to avoid drivetrain wind-up in four-wheel drive. KAM541 covers the front axle at 4.1:1; you'll need the matching KAM542 for the rear.
